摘要
在原位杂交研究的基础上,本文对慢性乙肝患者胞浆型HBV DNA及肝内其它乙肝指标进行综合分析。发现胞浆型HBV DNA可分3种类型:①肝内同时有核衣壳蛋白(HBcAg)与包膜蛋白(HBsAg)表达,此与HBV复制的产物表达特征一致;②肝内仅有HBsAg表达,此与HBVDNA和宿主基因组整合后的表达特点相似;③肝内HBsAg、HBcAg表达均缺如,可能代表一种HBV非复制状态。由此表明在HBV非复制期,出现胞浆HBV DNA杂交信号可能系DNA RNA杂交体所致。
Intrahepatocellular HBV DNA was analyzed in patients with chronic hepatitis B, by in situ hybridization assay, employing biotin labelled HBV DNA as the specific probe. It was found that HBV DNA was predominantely visualized in cytoplasma of the hepatocytes. In general, the patients with cytoplasmic HBV DNA could be divided into three groups: 1. those having at once intrahepatic HBsAg envelop protein and nucleocapsidated component (HBcAg) expression (this pattern of cytoplasmic HBV DNA could be replicative). 2. those having intrahepatic single HBsAg expression only (this observation was similar to the expression which happened after HBV DNA had been integrated into host genome); 3. those having no intrahepatic HBsAg and HBcAg expression in the liver. We speculate that the reason for cytoplasmic hybridization single in patients with non-replicative HBV infection may be DNA RNA transcript hybrid.
