摘要
目的 探讨氧自由基在肝脏保存再灌注损伤 (HPRI)中的作用及FK5 0 6的保护作用。方法 应用离体大鼠肝脏保存再灌注模型 ,观察肝组织超氧化物歧化酶 (SOD)活性 ,丙二醛 (MDA)含量 ,灌注液天冬氨酸转氨酶 (AST)活性 ,肝细胞形态学变化以及他罗利姆 (FK5 0 6 )对上述指标的影响。结果 HPRI时 ,SOD活性显著下降 (P <0 0 5 ) ,AST活性和MDA含量明显升高 (均P <0 0 5 ) ,肝细胞形态发生异常变化 ;使用FK5 0 6后 ,上述指标的异常变化均明显减轻 ,其差异有显著性 (P <0 0 5 )。结论 氧自由基是导致HPRI的重要因素 ,FK5 0 6能清除氧自由基 ,可以减轻HPRI。
Objective To explore the role of oxygen free radicals (OFR) in hepatic preservation reperfusion injury (HPRI) of rats and protective effect of tacrolimus (FK506).Methods SOD,MDA,AST and morphological changes of hypatocytes and effects of FK506 on them were observed during preservation reperfusion of isolated rat liver.Results Compared with the normal group,SOD activity decreased significantly (P<0.05),MDA and AST concentration increased remarkably (P<0.05),and abnormal morphological changes of hepatocytes were obvious during HPRI.In FK506-treated group,levels of MDA and AST were remarkably lowered (P<0.05),and SOP was significantly higher (P<0.05),compared with the control group.Conclusion It is indicated that OFR may be one of the primary causes for HPRI, and FK506 can attenuate HPRI by eliminating OFR.
