摘要
目的 :初步探讨P物质对体外培养人牙周膜成纤维细胞的作用与机制 ,以期为阐明正畸牙周组织改建中神经调控的机理提供依据。方法 :采用细胞培养和流式细胞技术 ,观察神经肽P物质及其受体拮抗剂对离体人牙周膜成纤维细胞DNA合成和细胞周期的影响。结果 :P物质使牙周膜成纤维细胞G1期细胞减少 ,S期细胞增加 ,促进更多的细胞进入增殖状态 ;P物质受体拮抗剂则明显抑制了P物质的作用。结论 :P物质对该细胞有促进增殖作用 ,并且可能是由SP受体 (NK1受体 )
Objective:To investigate the roles and mechanisms of SP on cultured human periodontal ligament fibroblasts.Methods:Effects of SP (10 -6 mol/L) and SP receptor antangonist (3x10 -5 mol/L) on DNA synthesis and cell cycle of human periodontal ligament fibroblasts were examined using cell culture and flow cytometer (FCM) techniques.Results:The number of S stage cells and multiplication index S+G 2M% markedly increased by SP( P <0.001);SP receptor antangonist(Spantide) inhibited it's effect( P <0.001) .Conclusions:SP may have proliferative effect on human periodontal ligament fibroblasts ,which might be mediated by SP receptor(NK 1 receptor).
