摘要 [摘 要]目的 研究适于大规模生产的流感病毒裂解剂、裂解和纯化方法。方法 使用不同表面活性剂(Triton X100和去氧胆酸钠)、不同浓度和时间进行裂解,通过电镜进行裂解效果观察。裂解后经密度梯度离心纯化,并进行抗原性试验。结果 Triton X100的裂解效果优于去氧胆酸钠,应用 1.0% Triton X100裂解 90min效果较好。裂解率达90%以上。裂解后纯化的样品抗原性与进口疫苗类似。结论 该工艺适于流感裂解疫苗大规模生产。 [Abstract] Objective To develop suitable lytic agent as well as lysis and purification procedure for large - scale production of influenza virus. Methods The influenza viruses were lysed with various surface active agents such as Triton X100 and sodium deoxycholate at different concentrations for different time,and the lysates were observed under electron microscope, purified by density gradient centrifugation, and detected for antigeniciry. Results An optimal procedure was developed by the lysis of influenza virus with 1.0% Triton X100 for 90min.The lysis rate of virus by the procedure reached above 90%, and the antigenicity of it was similar to that of imported vaccine. Conclusion The procedure is suitable for the large - scale production of vaccine by lysis of influenza virus.
