流式细胞术在血小板因子4保护环磷酰胺损伤小鼠骨髓中的应用价值 被引量:4

摘要 目的 探讨流式细胞术在研究血小板因子 4 (PF 4 )保护环磷酰胺 (CTX)损伤小鼠骨髓造血细胞中的应用价值。方法 腹腔一次性注射CTX 2 0 0mg/kg ,造成药物损伤模型。给CTX前予以PF 4预处理 ,于给药后第 5天、第 8天分别用流式细胞仪测定骨髓有核细胞DNA含量及细胞周期变化。结果 第 5天PF 4保护组的骨髓有核细胞中 ,G0 /G1期细胞百分率较CTX组明显提高 ,坏死、凋亡率下降 (P <0 0 1) ,而与对照组基本相同 (P >0 0 5 )。第 8天各组间比较无明显差异 (P >0 0 5 )。结论 PF 4对CTX损伤的小鼠骨髓造血细胞有保护作用。通过流式细胞仪测定DNA含量及细胞周期变化可阐明其化疗保护作用机制 ,表明流式细胞术具有重要的应用价值。 Objective To evaluate the use of flow cytometry in studying the chemoprotective effects of platelet factor 4(PF 4) on murine hematopoietic cells treated with cyclophosphamide (CTX).Methods The mice were intraperitoneally injected with PF 4 (40 μg/kg) twice at 6 hours intervals.20 hours after the second injection,they were given one injection of CTX(200 mg/kg) to develop models of chemical lesions.5 days and 8 days later,the DNA contents in bone marrow cells were detected by flow cytometry.Results 5 days after the use of CTX,the bone marrow cells in G 0/G 1 phase in PF 4-treated groups were found in significantly higher percentage than those in CTX-treated groups.The necrotic and apoptotic percentage were significantly lower,but there was no difference between the PF 4 groups and the control groups.Conclusions PF 4 can protect murine bone marrow from the cytotoxic effects of CTX.Detecting the DNA contents and cell cycle distribution by flow cytometry can demonstrate the effects and mechanism of PF 4,suggesting that flow cytometry is valuable for the study.
