P物质对下丘脑-腺垂体-睾丸轴调控作用的研究 被引量:3

摘要 目的 研究P物质 (Substancepower ,SP)对下丘脑、腺垂体、睾丸细胞分泌促性腺激素释放激素 (Go nadotropin -releasinghormone,GnRH)、黄体生成素 (Luteinizinghormone ,LH)、睾酮 (Testosterone ,T)的影响。方法 应用细胞培养技术 ,放射免疫测定法及下丘脑 -腺垂体 -睾丸三级培养细胞顺序灌注法。采用下列顺序灌注 ,SP浓度为 10 0 0ng/ 10 μl,( 1)H -AP ;( 2 )H (SP) -AP ;( 3 )H -AP (SP) ;( 4 )AP -T ;( 5 )AP-T (SP)。结果  ( 1)第 2组腺垂体细胞培养液LH含量比第 1组明显增加 ,二者比较 (t -test) ,P <0 0 1,差异有高度的显著性。 ( 2 )第 3组腺垂体细胞培养液LH含量与第 1组相比 ,P >0 0 5 ,二者之间无显著性差异。 ( 3 )第 5组睾丸细胞培养液T的含量比第 4组明显减少 ,二者比较 (t -test) ,P <0 0 1,差异有高度显著性。结论 ( 1)提示 :SP在下丘脑水平可能促进GnRH的分泌 ,从而促进腺垂体LH的分泌。 ( 2 )SP在腺垂体水平对LH的分泌无影响。 ( 3 )提示 :SP可能在睾丸水平发挥直接抑制作用。 Objective to investigate the effects of SP in the hypothalamus-pituitary-testis axis.Methods we used such methods as cell culture,radioimmunoassay and sequential perfusion of cell culture medium in adult male rats.Perfuse as follows(concentration of SP is 1000ng per 10μl):(1)H-AP,(2)H(SP)-AP,(3)H-AP(SP),(4)AP-T,(5)AP-T(SP).Results (1)The contents of Luteinizing hormone in the second group obviously increase compared to the first group.T-test showed that there were significant difference between two groups ( P <0.01) (2)The contents of Luteinizing hormone in the third group were not difference comparing with the first group.T-test showed that there were no significant difference between two groups ( P >0.05).(3)The contents of Testosterone in the fifth group were obviously lower than that in the fourth group.T-test showed that there were significant difference between two groups ( P <0.01).Conclusion 1.the results showed that SP may play a stimulatory role at the hypothalamus level,which resulted in a release of LH from AP cell cultures.2.SP can not significant effect on the release of LH in cultures of AP cells.3 the results suggested that SP may play an inhibitory role on the secretion of testosterone at the testis level.
