摘要
灵芝 (Ganodermalucidum)加硒深层培养后的菌丝 (含Se 16 0 0 μg/ g)分别经水及碱水提取、醇析、透析、Sevage法脱蛋白、DEAE cellulose柱层析纯化 ,得SeGLP 1。经SephadexG 10 0、聚丙烯酰胺凝胶电泳和紫外光谱分析鉴定 ,SeGLP 1为均一级分。GC与TLC分析表明 ,SeGLP 1含Gal、Glu、Xyl、Rha、Man ,其单糖摩尔比为Gal∶Glu∶Xyl∶Rha∶Man =0 .2 3∶1.0 0∶0 .72∶ 0 .2 5∶ 0 .0 9。SeGLP 1硒含量 16 42 μg/g。SeGLP 1中Se活性中心的结构可能为O =Se =O ,即Se取代了灵芝多糖GLP 1中 -OCH3结构上的CH3。
The polysaccharide containing selenium in Ganoderma lucidum (SeGLP 1) was isolated through water, lye and alcohol extraction ,dialysis, deproteining with Sevage method and purifying by chromatography of DEAE cellulose column. And SeGLP 1 was identified to be a homogeneous fraction with Sephadex G 100 and polyacrylamide gel electrophoresis and UV spectrum analysis, while Gal, Glu, Xyl, Rha, Man were determined to be components of SeGLP 1 by GC and TLC. The quantitative analysis of SeGLP 1 showed that the molar ratio of Gal∶Glu∶Xyl∶Rha∶Man=0.23∶1.00∶0.72∶0.25∶0.09, and that SeGLP 1 contained 1642μg/g selenium. And the results from IR, 77 Se NMR, 13 C NMR and Laser Raman spectra showed that the structure of activity center of selenium of SeGLP 1 might be O=Se=O,and CH 3 in -OCH 3 group of GLP 1 was substituted by selenium forming this structure.
