摘要
目的 :探讨多胺生物合成抑制剂二氟甲基鸟氨酸 (α- difluoeom ethyl- ornithine,DFMO)抗人膀胱癌 EJ细胞增殖的作用。方法 :活细胞显微摄影、伊红染色细胞计数、绘制细胞生长曲线方法。结果 :4m mol~ 8mmol DFMO可以抑制人膀胱癌 EJ细胞的增殖 ,并具有剂量依赖性 ;外源性腐胺可以阻止 DFMO上述作用。结论 :DFMO抗人膀胱癌 EJ细胞增殖的作用是通过抑制多胺生物合成而实现的 ;抑制多胺生物合成可能是治疗膀胱瘤的一种有效途径。
Objective:To observe the effect of DFMO,an inhibitor of ODC-a key enzyme of polymine biosynthesis,on the proliferation of EJ cells of bladder carcinomas.Methods:Micrography of Living Cells.Diagram of Cellular Growth.Results:4~8 mmol DFMO can induce EJ cells proliferation arrested.It was related to the dose that DFMO acted.These changes resulted from DFMO were prevented,however,when the cells were treated by DFMO along with exogenous putresine.Conclusion:DFMO arrests the proliferation of EJ cells by preventing ploymine biosynthesis.The control of polyamine biosynthesis may be an effective approach for bladder tumor therapy.
