摘要
目的 :探讨高浓度葡萄糖对人腹膜间皮细胞 c- myc和 p2 1基因表达的影响。方法 :体外培养的人腹膜间皮细胞经含 1.5 %、2 .5 %、4 .2 5 %葡萄糖的 M199培养基培养 2 4 h,以正常 M199培养基和含 4 .2 5 %甘露醇的 M199培养基为对照。应用半定量 RT- PCR检测 c- m yc、p2 1基因 m RNA表达。结果 :2 .5 %葡萄糖组、4 .2 5 %葡萄糖组间皮细胞 c- myc和 p2 1m RNA的表达量显著高于 1.5 %葡萄糖组、M199对照组及 4 .2 5 %甘露醇渗透压对照组 (P<0 .0 5 ) ;而 1.5 %葡萄糖组、M199对照组及 4 .2 5 %甘露醇渗透压对照组间间皮细胞 c- myc和 p2 1m RNA的表达量无显著差异 (P>0 .0 5 ) ,且葡萄糖浓度越高 ,c- myc和 p2 1m RNA表达量越高 (r=0 .96 8,0 .96 7,P<0 .0 1)。结论 :高浓度葡萄糖能上调 C- myc和 P2 1m RNA基因表达 ,这种作用与葡萄糖浓度呈正相关。
Objective:To investigate the effect of glucose on expression of c-myc and P21 gene in human peritoneal mesothelial cells.Methods:The cultured human mesothelial cells were exposed to culture medium containing different concentrations of glucose (1.5%,2.5%,4.25%),4.25% mannitol and normal culture medium as control for 24 hours,then mRNA expression was measured by semi-quantitative reverse-transcript PCR.Results:The level of c-myc mRNA and P21 mRNA expression was significantly higher in 2.5% glucose group and 4.25% glucose group than in 1.5% glucose group,4.25% mannitol group and normal culture medium group (P<0.05).But there was no significant difference between the 1.5% glucose group,4.25% mannitol group and normal culture medium group(P>0.05).The level of c-myc and P21 mRNA expression incresed as glucose concentration increased(r=0.968,0.967,P<0.01).Conclusion:High glucose can up-regulate c-myc and P21 gene expression in a concentration-dependent manner.
