GM-CSF对膀胱癌EJ细胞株CD44v6表达的影响

摘要 目的 :明确GM -CSF对EJ细胞增殖及对CD44v6表达的影响。方法 :MTT法检测GM -CSF和IFN对EJ细胞增殖的影响 ,免疫细胞化学法观察GM -CSF对CD44v6表达的影响。结果 :GM -CSF以不同浓度作用EJ细胞对其增殖无影响 ,但GM -CSF联用IFN可抑制EJ细胞增殖 ,GM -CSF连用IFN可降低CD44v6表达。结论 :GM -CSF +IFN可降低CD44v6在EJ细胞中的表达。 Objective:To determine whether the expression of CD44v6 in bladder cancer is associated with tumor malignant behavior and to investigate whether GM-CSF can affect proliferation and expression of CD44v6 of EJ cell line. Methods: EJ cell line was cultured, and proliferation and CD44v6 expression were tested by the MTT method or immunocytochemistry method respectively in the presence of different concentrations of GM-CSF alone or combined with IFN-γ. Results: Compared with control, proliferation of human bladder cancer EJ line treated with GM-CSF alone was not changed, but it was significantly inhibited if treated simultanuously with GM-CSF and IFN-γ. Using the monoclonal antibody to CD44v6, it was found that expression of CD44v6 in human bladder cancer EJ cell line was markedly dereased after GM-CSF and IFN-γ treatment for 48 hours by immunocytochemistry method. Conclusion: Our study indicated that CD44v6 may play an important role in human bladder cancer progression and metastasis, and CD44v6 seems to be a marker of high metastasis potential of bladder cancer. Combination treatment of GM-CSF and IFN-γ may downregulate CD44v6 expression of EJ cell line.
