摘要 目的 观察DPC4/Smad4 mRNA在胰腺癌组织中的表达及其临床意义。方法 应用原位杂交技术对23例胰腺癌组织中的DPC4/Smad4 mRNA表达进行研究。结果 胰腺癌组织中DPC4/Smad4 mRNA表达率为52.2%(12/23),低于癌周组织表达率9/10。统计学分析显示,DPC4/Smad4mRNA表达与胰腺癌病理分级呈显著相关(P<0.05),与胰腺癌临床分期及有无局部淋巴结转移间无显著相关(P>0.05)。结论 DPC4/Smad4 mRNA基因表达有可能作为胰腺癌诊断与鉴别诊断的生物学指标之一。 Objective The aim of the study was to observe the expression of DPC4/Smad4 mRNA in pancreatic carcinoma and to analyse the relationship between expression of DPC4/Smad4 mRNA and clinical manifestations. Methods The expression of DPC4/Smad4 mRNA in pancreatic carcinoma and pericancerous tissue were detected by in situ hybridization method. Results The positive rates of DPC4 mRNA in pancreatic carcinoma and pericancerous tissue were 52. 2% (12/23) and 9/10 respectively. The expression of DPC4 mRNA was related to the degree of cell differentiation of pancreatic carcinoma, but no relation was found between the DPC4 mRNA expression and the clinical stage or the status of lymph node metastasis. Conclusions The results suggest that DPC4/ Smad4 is an important tumor suppressive gene that is frequently deletable in pancreatic carcinoma. Inactivation of DPC4 may play a role in the carcinogenesis of pancreatic carcinoma.
