摘要
目的建立阻塞性黄疸大鼠模型,从血清蛋白基因转录水平观察肠外营养(PN)对改善阻塞性黄疸动物营养不良的有效性。方法SD大鼠36只,随机分为4组,每组9只,两组行假手术(SHAM),其中一组为经口饮食对照组,另一组行PN;另两组行胆总管结扎术(CBDL),其中一组为经口饮食对照组,另一组行PN。在PN前、PN72及120小时后,检测其血清白蛋白水平、肝脏白蛋白(ALB)mRNA和白蛋白转录因子D-位点结合蛋白(DBP)mRNA水平。结果CBDL大鼠PN120小时后血清Alb水平略有上升;所有PN大鼠肝脏ALBmRNA和DBPmRNA水平均高于经口饮食对照组。同时发现,SHAM大鼠血清白蛋白水平与肝脏ALBmRNA间、肝脏DBPmRNA间、肝脏ALBmRNA与DBPmRNA水平显著相关;而在CBDL大鼠中这些指标间无显著相关性。结论PN可显著提高DBP转录,后者可有效启动白蛋白基因的转录,但可能受其他因素影响。合理配方的PN能有效维持CBDL大鼠血清白蛋白水平。
Objective This study detected serum albumin level,hepatic albumin mRNA and D-site bin d-ing protein mRNA levels of obstructive jaundice rats at PN0h,72h,and120h respectively,to in vestigate the efficiency of PN to improve obstructive jaundice rats' malnutrition.Methods 36SD rats were divided into4groups randomly(n=9).SHAM operation was performed in2groups,one of which was assigned to the oral diet control group,and the other to PN group.Common bile duct ligation(CBDL)was performed in2groups,one of which was assigned to oral diet control group,and the other to PN group.Results After120h PN,serum albumin level of CBDL rats slightly rose,and PN rats' Alb mRNA and DBP mRNA levels were higher than those of oral diet control group.There was a significant correlation between serum albumin and hepatic Alb mRNA level,serum albumin and hepatic DBP mRNA level,hepatic Alb mRNA and DBP mRNA level in the SHAM rats,while there wasn' t an significant correlation between the indices in the CBDL rats.Conclusions PN was an effective method to maintain rat serum albumin level and DBP could effectively startup transcription of albumin,but there was no significant correlation between DBP and serum albumin levels.
