一种快速测定Ks的新方法

摘要 该研究建立了以生物传感为基础的细胞呼吸初速度测定法,用于确定Ks。实验采用动态模型,微生物传感器响应斜率R=dI/dt,响应值表示微生物对代谢底物的相对外源呼吸速率,通过双倒数作图法求出Ks。用本法测得Pseudomrnoas sp.A_4,E.coli N/B和Sacchromyces cerevisiae氧化葡萄糖的Ks分别为3mg/L.7mg/L和135rag/L。Ps.sp.A_4氧化苯酚的Ks为1.5mg/L,该方法具有快速、简便、直观和重现性好等特点,有一定实用意义。 A novel way for Primary determination of microbial Ks has been developed based on of biosensor method. Viable cells of selected species of microbial organisms were hold in narrow space between oxygeu probes and porous membranes(0.3μm pore size) to form microbial sensors. Using the devices, the exorespiration rates on a series of concentrations of substrates could be estimated in real time and expressed as the responce slopes those were determined according to rinetic responce model R=dI/dt, followed by caculation of Ks from Lineweaver-burk plot. The features of the method has been discussed in this article.
