淫羊藿甙对体外培养成骨细胞增殖和分化的影响

The effect of icariin on prolifetration and differentiation of osteoblasts in vitro

目的探讨淫羊藿甙对体外培养成骨细胞增殖和分化的影响及其作用机制。方法采用体外成骨细胞培养技术,进行细胞计数和碱性磷酸酶活性测定,观察淫羊藿甙对成骨细胞生长增殖和分化的影响。结果(1)不同浓度的淫羊藿甙对成骨细胞的生长增殖均有促进作用,以10ng/ml浓度时其作用最强(P<0.05);(2)淫羊藿甙可明显抑制分化早期成骨细胞内ALP活性,而对分化晚期成骨细胞内ALP活性具有促进作用;(3)淫羊藿甙对成骨细胞的作用与药物作用时间有关。结论淫羊藿甙促进成骨细胞增殖的同时,增强了成骨细胞活性。

Objective To demonstrate the effect of icariin on proliferation and differentiation of osteoblasts in vitro.Methods Calvaria of the newbone Wistar rat was used and th e osteoblast culture technique was a dopted for the study.The effect of ic ariin on proliferation and differentiation of osteoblastes in vitro was observed by cell count and a ssay of ALP activity.Results(1)Various doses of icariin could affect the osteoblast proliferation.Icariin was the most effective with the concentration of 10ng /ml (P<0.05).(2)Icariin might depress ALP activity in the early stage of the cultured osteoblasts and improve ALP activity in the later stage.(3)The results demonstrated that the ic ariin could improve the osteoblastic prolifera tion in the early stage,and had more b enefit for the differentiation of th e osteoblasts in the later peri-od.Conclusion The icariin could improve osteoblastic proliferation and develop the osteoblastic activity .
