摘要
将猪囊虫抗原基因 TS76的真核表达型质粒 VTS76单独或与 p UC18联合肌肉免疫注射于 BAL B/ c小鼠 ,以MTT比色法检测小鼠脾淋巴细胞 Con A刺激的增殖反应及 IL - 2的诱生活性 ,EL ISA方法检测免疫小鼠 Ig G总量和特异性抗体水平 ,常规法检测外周血免疫细胞数量的动态变化。结果发现 ,VTS76免疫小鼠各项细胞和体液免疫应答反应指数均比空白对照组小鼠显著提高 ;联合免疫组小鼠的细胞免疫应答和体液免疫应答反应指数均比 VTS76小鼠提高。由此可见 ,以 VTS76免疫小鼠可诱导其特异性细胞和体液免疫反应 ,而 p UC18又明显提高了 VTS76免疫小鼠的免疫应答水平 。
Sixty BALB/c mice of 2 months old were randomly divided into three groups and twice inoculated intramuscularly with different plasmids at the interval of 14 days.Group A was injected with VTS76,a recombinant plasmid of an antigenetic gene of Cysticercus cellulosae.Group B were injected with VTS76 and pUC18.Group C was injected with VR1020,a eukaryotic expression plasmid,as control group.Mice were bled on 0,7,14,21,28 and 35 days after immunization.The immune cells of mice were accounted as the routine method.The amount of IgG and the titres of specific antibody of immunized mice were determined by ELISA assay.The proliferative reaction of lymphocytes with ConA was detected;the bioactivity of interleukin-2 induced by ConA was investigated by MTT assay.The experimental results showed that the amounts of total IgG and specific antibody of the immunized mice were higher than that of control group.The level of induced interleukin-2 bioactivity significantly increased in the spleen cells from immunized mice.Lymphocytes,macrophages and neutrophils of immunized mice remarkably elavated in comparison with those of the control.These indicate that VTS76 can effectively raise the immune responses of BALB/c mice against Cysticercus cellulosae.The coinoculation of the immunized mice with pUC18 elicited stronger humoral and cellular immune responses as compared with those of the immunized mice.So TS76 gene could be employed as a putative immunoprotective gene in the further development of DNA vaccine against swine cysticerosis,and pUC18 could be used as immunoenhancing adjuvant for DNA vaccine.
