盐酸克林霉素(clindamycin)在健康猪及链球菌感染猪体内的药代动力学

Pharmacokinetics Hydrochloride of Clindamycin in Swines

盐酸克林霉素按每公斤体重 11mg单一剂量肌肉注射 ,分别观察了其在健康猪 (n=5 )和链球菌感染猪 (n =5 )体内的药代动力学。用高效液相色谱法测定盐酸克林霉素的血药浓度 ,所得血药浓度 -时间数据采用 MCPKP软件进行分析。在健康猪体内的药代动力学参数如下 :Cmax为 (2 .5 46 43± 0 .2 2 42 ) mg/ L,T1 /2 为 (3.2 10± 0 .795 3) h,AUC为(17.86 5 72± 1.2 5 46 ) m g· h/ L ,Tmax为 (2 .18796± 0 .44 37) h。在链球菌感染猪体内的药代动力学参数如下 :Cmax为(2 .5 5 6 89± 0 .12 45 ) mg/ L ,T1 /2 为 (2 .6 912 3± 0 .8895 2 ) h,AUC为 (12 .0 0 6 5 4± 3.2 5 46 ) mg· h/ L ,Tmax为(1.16 176± 0 .5 436 ) h。

In order to prepare a contagious disease model for pharmacokinetic studies in swine,5 hybrid pigs were used to develop a typical acute streptococcosis suum by inoculating subcutaneously 0.35 billion of pure living Streptococcus suis (C55121) to each animal for per weight. A plasma sample (1 mL) was pipetted into Oasis TMHLB spiked with 10 μL of internal standard,and loaded onto the silica extraction column. Each extraction column was preconditioned with one volume of methanol (about 1 mL) and one volume of deionized water (about 1 mL). An aliquot (20 μL ) was injected for HPLC analysis. Elution was achieved at ambient temperature with acetonitrile, tetrahydrofuran and 0.05 mol/L phosphate bulfer as the mobile phase. The flow-rate was set to 1.0 mL/min, UV-detector was operated at 205 nm. The pharmacokinetic of clindamycin was carried out in 5 healthy pigs and 5 infected pigs by single IM administration at the dose of 11 mg/kg body weight and the plasma concentration of clindamycin were determined in the above-mentioned conditions. The main pharmacokinetics parameters after IM administration were as follows: in healthy pigs, C max(2.546 43±0.224 2) mg/L, T 1/2(3.210±0.795 3) h,AUC(17.865 72±1.254 6) mg·h/L,T max(2.187 96±0.443 7) h. In infected pigs, C max(2.556 89±0.124 5) mg/L, T 1/2(2.691 23±0.889 52) h, AUC(12.006 54±3.254 6) mg·h/L, T max(1.161 76±0.543 6) h. So when a streptococcosis disease be treated we should use a dose of 20 mg/kg body weight.
