摘要
建立了鸡血浆和肌肉中替硝唑 (Tinidazole,TNZ)的高效液相色谱 (HPL C)测定法。采用 Shim adzu L C- 6 A型高效液相色谱系统 ,YWG- C1 8柱 ,UV检测器 ,以甲醇 /水 (32∶ 6 8)为流动相 ,流速 0 .8m L/ m in,波长 318nm,室温测定。用二氯甲烷萃取血浆和肌肉中 TNZ。结果表明 ,TNZ的最低检出限为 0 .0 12 5 mg/ L,在血浆和肌肉中的最低定量限分别为 0 .0 2 5 mg/ L和 0 .0 2 5μg/ g。其质量浓度在 0 .0 2 5~ 6 4mg/ L范围内与色谱峰高呈良好线性关系 (r=0 .9999)。TNZ质量浓度为 0 .0 2 5、1.6、6 4mg/ L 时 ,日内相对标准差小于 5 % ,日间相对标准差小于 10 % ,在血浆和肌肉中的回收率分别为 98.0 0 %、91.88%、96 .17%和 90 .0 0 %、99.0 6 %、92 .5 4%。该方法灵敏、快速、简便 ,适用于
A high performance liquid chromatographic(HPLC) method was developed for determination of tinidazole(TNZ) in plasma and tissues of chickens.In the shimadzu LC-6A HPLC system a YWG-C 18 column and UV detertor at 318 nm wavelength was used.The mobile phase was methanol-water(326∶8 v/v) with the flow of velocity 0.8mL/min.TNZ in plasma and muscle tissues extracted with dichloromethane.The result showed that:the minimal limit of detection and the minimal limit of quantitative of TNZ was 0.012 5 ug/mL and 0.025 μg/mL plasma(0.025 μg/g tissues),respectively.A good linear response was obtained for TNZ standard from 0.025 μg/mL to 64 μg/mL with the correlation coefficient to be 0.999 9.When concentration of TNZ was 0.025,1.6,64 μg/mL,the RSD in within-day and between-day was less than 5% and 10%,respectively.The TNZ recovery from plasma and muscle tissues was 98.00,91.88,96.17% and 90.00,99.06,92.54%,respectively.The method was proved to be sensitive,rapid,simple and convenient.It may be applied to study of pharmacokinetics and determination of residues of TNZ in chickens.
