摘要
以从 (5 0~ 6 0℃ )酸性温泉中采集的菌为选育对象 ,研究确定浸矿高温菌初筛、复筛的方法后 ,用该方法获得高温浸矿菌retech Y。通过生理生化的研究鉴定retech Y为兼性异养菌 ,此高温菌的最适生长条件为 :FeTSB0 培养基 ,pH 1.6 0 ,Fe2 + 含量 90mmol/L ,最适温度为 6 0℃ ,最适生长周期为 2 4h ,Fe2 + 氧化率为 10 0 % ,菌数为 7× 10 7个 /ml。
An screening assay was developed in order to directly and simply isolate moderately thermophilic and acidopHilic iron oxidizing Thiobacillu from acidophilic geothermal areas. This method was used to obtain a thermophilic chemolithoheterotrophs strain named as retech Y from acidopHilic hot spring. The isolated species were grown in ferrous iron yeast extract medium containing 90 mmol/L ferric iron, 0.035% yeast extract, and basal salt, in which iron acts as the energy source and yeast extract is the carbon source.The results show that the optimal temperature is 60℃ and pH value is 1.60 for its growth condition. When incubated for up to 24 h, the number of 7×10 7 cell/ml is obtained and 100% ferric iron is oxidized.
