组织工程用大鼠肌卫星细胞的体外筛选 被引量:12

摘要 目的 探讨不同年龄肌卫星细胞体外培养增殖、分化规律 ,以获得增殖能力强、分化能力适度的组织工程用卫星细胞群。 方法 用MTT法、免疫细胞化学技术及粘附细胞仪等手段 ,检测不同年龄段大鼠多代肌卫星细胞的增殖、分化能力和细胞数量。 结果  (1)单位重量细胞数与动物年龄成负相关 ;(2 )不同年龄组间细胞增殖能力从 7个月龄始明显递减 ;(3 )粘附细胞仪检测细胞分化能力 :2~ 3个月龄细胞传 12~ 13代后Desmin单抗荧光强度明显减弱。 8个月龄则是 7~ 9代后明显减弱。 结论 肌组织工程需要的卫星细胞体外扩增的临界传代数 :6个月龄以下的大鼠为 10~ 12代 ,7个月龄以上的大鼠为 5~ 7代。 Objective To find out of proliferation and differentiation about satellite cells (SCs) from various age rats in vitro, and explore the best extent of proliferation for muscle tissue engineering Methods The quantity and quality of SCs from various age rats were compared by MTT, Immune cytochemistry and ACAS Results (1)The number of SCs was a negative correlation with the age of animal (2)It was very stable of the activity activity and growing ability of SCs from age of 1 month to 6 month, but after 7 months old it becomes worse (3)The ability of differentiation was tested by ACAS, fluorescence intensity of Desmin antibody became weakness from 12~13 passage for 2~3 months old, 7~9 passage for 8 months old when passages are growing for 2~3 month old and 8 monthe old groups The isolated cells were idntified to be Scs, and Myotubes were present finally; Cnoclusions The passages of muscle tissue engineering is 10~12 passage less than 6 months old, and 5~7 passage over 7 months old
