摘要 目的 观察机械张力对人成骨细胞TGF - β基因表达的影响。 方法 通过Flexercell细胞拉伸力学装置对人成骨样细胞MG - 6 3进行 6 %、12 %和 2 4 %的拉伸应变加载实验 ,用RT -PCR反应检测细胞受力后TGF - βmRNA的表达。结果  6 %和 12 %的拉伸率能显著促进MG - 6 3细胞TGF - β的表达 ,但 2 4 %的拉伸率作用对细胞TGF - β基因表达影响不明显。结论 恰当大小的机械张力可以增加成骨细胞TGF - β的表达 ,过大的应力刺激可能没有这种效应。 Objective To study the effects of mechanical strain on the expression of TGF-β in human osteoblasts.Methods Cloned MG-63 human osteoblasts were cultured,and seeded at 10 5/well on 6-well Bioflex cell plate.Subsequently,the cells were exposed to mechanical strain stimulation(6%,12% and 24% elongation rate) using Flexercell 3000 Cell Stretching Unit.RT-PCR reaction was used to assess the TGF-β mRNA expression in MG-63 osteoblasts.Results mRNA for TGF-β 1 increased significantly in the strained MG-63 cells with 6% and 12% mechanical streching,24% stretching showed little effect on the mRNA expression of TGF-β 1.Conclusion This study suggests that proper mechanical strain increase the expression of TGF-β in osteoblasts.However,overloading may not have this effects.
