摘要 目的 :研究巨噬细胞毒素降低反复自然流产模型CBA/J×DBA/ 2小鼠胚胎吸收率的免疫机制 .方法 :在小鼠交配前腹腔注射变性蛋白质包被的二氧化硅颗粒 ,做为巨噬细胞毒素以去除巨噬细胞 ,观察胚胎吸收率和淋巴细胞亚群变化 ,进而分析淋巴细胞浸润与妊娠结局的关系 .结果 :二氧化硅预处理组平均每窝活胎数显著增多 (6 3± 1 5vs 3 5± 1 0 ,P <0 0 1) ,胚胎吸收率则显著下降 (18 2 %vs .38 2 %,P <0 0 5 ) .与此相应 ,CD3+ CD6 9+ 、CD3+ CD71+ 细胞比率和CD4 5 +MHC -II+ 、CD80 + MHC -II+ 细胞比率均显著降低 .结论 :体内注射巨噬细胞毒素可以显著降低CBA/J×DBA/ 2小鼠母胎交界面活化型T细胞和某些巨噬细胞亚型的比率 ,并且这种变化与胚胎吸收率的下降相联系 . Aim: To investigate the immunological mechanisms for the reduction of embryo resorption with macrophage toxin in CBA/J×DBA/2 mice, as a model of recurrent spontaneous abortion. Methods: Murine macrophage depletion was performed by i.p. injection of denatured protein-coated silica particle as a macrophage toxin before mating. The resorption rate of embryos was calculated and the proportions of lymphocyte subpopulation were observed in order to determine the relationship of lymphocyte infiltration and pregnancy outcome. Results: The number of viable fetuses per litter in CBA/J×DBA/2 mice pretreated with silica was significantly higher compared to control group (6 3±1 5 vs. 3 5±1 0, P<0 01), whereas the resorption rate of embryos was significantly lower (18 2% vs. 38 2%, P<0 05). Accordingly, lower proportions of CD3 +CD69 +, CD3 +CD71 + cells and CD45 +MHC-II +, CD80 +MHC-II + cells were associated with silica preteatment. Conclusion: In vivo depletion of macrophages by silica pretreatment could reduce embryo resorption in CBA/J×DBA/2 mice, which was associated with the decrease of activated T cell proportions and some subsets of macrophages.
