复方中药诱导Burkitt′s淋巴瘤Raji细胞凋亡作用的研究

STUDY ON TRADITIONAL CHINESE MEDICINE RECIPE IN INDUCING BURKITT′S LYMPHOMA CELLS-RAJI CELLS APOPTOSIS

目的 :研究复方中药对 Burkitt′s淋巴瘤细胞株 Raji细胞的影响 ,并探讨其机制。方法 :采用 MTT法检测细胞毒作用 ,利用 Wright- Gimesa染色法 ,Hoechst 33342荧光染色等手段观察凋亡细胞的形态结构变化 ,应用透射电镜技术和流式细胞分光光度术进行凋亡定性定量观察 ,并探讨凋亡抑制基因 bcl- 2蛋白表达。结果 :复方中药对 Raji细胞具有明显的生长抑制作用。共同孵育一段时间后 ,细胞呈现典型的凋亡形态学变化。流式细胞仪分析表明复方中药能使 Raji细胞发生凋亡 ,凋亡率呈现对浓度和时间的依赖性 ,并发现复方中药使 Raji细胞 bcl- 2基因的表达下调。结论 :复方中药能诱导 Raji细胞发生凋亡 ,其作用机制与 bcl- 2基因表达的下调有关。

Objective:To study the effect and mechanism of CTM recipe in inducing the human B lymphoma cell line Raji.Methods:MTT,light microscopy,transmission electromicroscopy (TEM) and flow cytometry were used.Results:The results showed that CTM recipe could inhibit the proliferation of Raji cells.The in vitro effect of CTM on the growth of Raji cells was evaluated by MTT assay.The IC 50 values were (0.492±0.093) micrograms/mL.The apoptosis of Raji cells was conformed by flow cytometry and characteristic ultrastructure alterations.The results also demonstrated that inhibitory effects of CTM recipe were associated with the high percentage of hypodiploid cells.The expression of bcl 2 protein decreased in a dose and time dependednt manner in Raji cells.Conclusion:It is suggested that induction of cell apoptosis can be one of the mechanisms of the therapeutic effect of the CTM recipe.Moreover,this apoptosis induction occurs through the modulation of bcl 2.
