摘要
目的 :探讨大鼠脊髓损伤后GDNFmRNA在坐骨神经的表达变化及其意义。方法 :Allen’s方法致大鼠T13段脊髓不完全损伤后 ,以 β Actin为内参照物 ,应用半定量RT PCR方法 ,观察大鼠坐骨神经在脊髓损伤前、后不同时间GDNFmRNA表达变化。结果 :脊髓损伤前GDNFmRNA在大鼠坐骨神经仅有微量表达 ,脊髓损伤后 2 4h表达增加 4倍 ,72h增加 15倍 ,7d增加 3倍 ,10d仍高于正常水平。结论 :脊髓损伤后坐骨神经高表达GDNFmRNA ,是神经元通过“胞体—轴突—靶器官”途径自我保护的一种反应形式 。
Objective: To investigate the gene expression of glia cell line derived neurotrophic factor GDNFmRNA on sciatic nerve after crushed spinal cord injury(SCI) in rats.Methods:The speciment of sciatic nerve was taken at different time before and after T 13 segments of spinal cord were injured by modified Allen crush method (10g×7.5cm).The level of GDNFmRNA was observed and compared before and after SCI,using semi quantitative RT PCR method with β Actin as an inner consult.Results:Much minim expression of GDNFmRNA was found in sciatic nerve of normal adult rats,but it increased 4 times 24 hours after SCI,15 times 72 hours,3 times 7 days and slight higher than normal level 10 days after SCI.Conclusion:It should be one kind of self protective behave models for sciatic nerve to express high standard GDNFmRNA through 'neuron axon target' route in the early stage after SCI.GDNF should be supplied early when it was applied to treat SCI.
