摘要
目的 系统观察烧伤血清对肠上皮细胞屏障功能的影响。 方法 制作体外大鼠肠上皮细胞IEC - 6株烧伤血清刺激模型 ,实验分为对照组和烧伤血清组 ,采用生化检测、图像分析、细胞ELISA、放射免疫等技术 ,动态观察肠上皮细胞活性、细胞Ca2 + 浓度、细胞通透性以及细胞骨架的变化。 结果 肠上皮细胞经烧伤血清作用后 ,细胞活性即开始降低 ,而乳酸脱氢酶升高 ,胞浆Ca2 + 浓度增高 2 .4 3倍 ,单层细胞通透性显著增加 ,细胞骨架成分肌动蛋白 (F -actin)、角蛋白(keratin)、微管蛋白 ( β-tubulin)表达减少或状态改变。 结论 烧伤血清刺激后肠上皮细胞活性降低 ,通透性增加 ,细胞骨架表达减少 。
Objective To study the effect of serum from burned animals on barrier function of intestinal epithelial cells (IEC). Methods Intestinal epithelial clones (IEC-6) stimulated with serum were used as in vitro models. The experiment consisted of 2 groups, the control group and the burned serum group. The changes of cytoactivity, the level of cellular Ca 2+ , the permeability of the IEC and the cytoskeleton were dynamically observed with image analysis, enzyme-labeled immunosorbent assay (ELISA) and radioimmunity technique. Results In the early stage after being stimulated by serum from burned rats, the cytoactivity obviously decreased. The content of lactate dehydrogenase (LDH), the level of cellular Ca 2+ , and the permeability of the IEC increased. And the content and expression of cytoskeleton markedly decreased. Conclusions Serum from burned rats can make the IEC cytoactive decrease, the IEC permeability increase and the expression of the IEC cytoskeleton decrease, so as to damage the barrier function of the intestinal mucosa.
