摘要 目的:探讨非甾体类抗炎药(nonsteroidal anti-inflammatory drugs,NSAID)对体外培养的胃癌细胞SGC7901生长及环氧合酶活性的作用。 方法:用MTF法观察阿司匹林、尼美舒利对胃癌细胞SGC7901生长的影响;免疫细胞化学方法检测Ki-67及COX-2蛋白表达;采用放射免疫分析法测定细胞培养上清PGE2水平。 结果:阿司匹林和尼美舒利呈时间、剂量依赖性方式抑制细胞增生,减少Ki-67蛋白表达。胃癌细胞SGC7901COX-2蛋白表达阳性,并伴有PGE_2释放,尼美舒利可减少PGE_2产生,但对COX-2蛋白表达无影响。 结论:阿司匹林和尼美舒利均可抑制COX-2表达阳性的胃癌细胞SGC7901生长,尼美舒利通过抑制COX-2酶活性而减少PGE_2释放,该研究提示COX-2可能在胃癌细胞SGC7901增生中起重要作用。 AIM: To evaluate the potential role of nonsteroidal anti-inflammatory drugs (NSAID) on the proliferation and cyclooxygenase (COX) activity of gastric adenocarcinoma cells SGC7901.METHODS: After treatment with aspirin and nimesulide, SGC7901 cell growth was quantified by MTT assay, and the expression of Ki-67 and COX-2 protein in the cells was detected by immunocytochemical staining. PGE2 level in the culture supernatant was measured by radioimmunoassay.RESULTS: Both aspirin and nimesulide inhibited the cells proliferation in a time and dose-dependent fashion, and the expression of Ki-67 was reduced COX-2 protein expression and PGE2 production were detected in SGC7901 cells. Nimesulide decreased the release of PGE2, whereas the expression of COX - 2 was unchanged.CONCLUSION: Aspirin and nimesulide can inhibit the growth of gastric cancer cells SGC7901 expressing COX-2 protein. Treatment of SGC7901 cells with nimesulide resulted in a reducing of Ki-67 expression and PGE2 release. These data indicate that COX-2 may play an important role in the proliferation of SGC7901 cells.
