摘要
目的:研究丹参酸乙(salvianolic acid B,SAB)的抗氧化作用对大鼠肝星状细胞(hepatic stallate cells,HSC)增生周期的影响。 方法:采用原位灌注法消化大鼠肝脏,108 g·^(-1)nycodenz密度梯度离心,分离肝星状细胞,分别以不同浓度的SAB或MDA/SAB处理细胞,~3H-TdR掺入法观察细胞的增生能力,流式细胞术分析细胞周期各时相变化及凋亡情况。 结果:1和10 μmol·L^(-1)SAB可显著抑制HSC的增生(2862±1234&2988±407 vs 4986±727,P<0.05,P<0.01),这种作用主要是抑制了细胞周期过程中G_1期向S期的过渡;1和10μmol·L^(-1)SAB均可抑制MDA刺激的HSC增生(3067±731&3042±321vs 4007±587,P<0.05,P<0.01);两组剂量的SAB均不促进HSC的凋亡。 结论:丹参酸乙可抑制体外培养HSC的增生,这种抑制作用与SAB的抗氧化作用有一定的关系。
AIM:To research the effects of anti-oxidation of salvianolic acid B (SAB) on proliferation of rat cultured hepatic stellate cells.METHODS: Hepatic stellate cells were derived from normal rat by perfusion and digestion in situ and density gradient centrifugation with 108 g · L-1 nycodenz. Then cells were treated by various concentrations of SAB or MDA/ SAB. Later, the proliferation of cells were observed by 3H-TdR incorporation; the changes of cell cycle and apoptosis were analyzed by flow cytcmetry.RESULTS: Both 1 μmol·L-1 and 10μmol·L-1 of SAB obviously suppressed the proliferation of hepatic stellate cells (2862 ± 1234& 2988 ± 407 vs 4986 ± 727, P < 0. 05 and P < 0. 01 respectively) by the arrest of the cell cycle processes from G1 phase to S phase; both concentrations of SAB suppressed the proliferation of MDA-stimulated hepatic stellate cells (3067 ± 731&3042 ± 321 vs 4007 ±587, P < 0.05 and P<0.01 respectively); and neither of them increased the apoptosis of hepatic stellate cells.CONCLUSION: SAB can inhibit the proliferation of rat cultured hepatic stellate cells, which is associated with the antioxidant effect of SAB.
