人肝癌细胞在免疫重建前后SCID小鼠中的生长特点

摘要 目的 探讨 SCID小鼠免疫重建及免疫对人肝癌细胞成瘤的影响 .方法 体外培养人肝癌细胞系 ,接种到 SCID小鼠和用 BAL B/ c,CBA/ N小鼠外周血淋巴细胞重建的SCID(B- PBL- SCID和 C- PBL- SCID)小鼠皮下 ;监测成瘤状态 ,测定鼠脾细胞毒杀伤力和免疫重建的免疫球蛋白量 ,作血清与靶细胞凝集实验 .结果 免疫重建体现了所用小鼠免疫对肿瘤的排斥特点 ,免疫重建成功 ;SCID和 C- PBL- SCID小鼠 10 0 %成瘤 (两瘤体相比 P<0 .0 1) ,B- PBL - SCID出现瘤体后逐渐消失 ;B- PBL- SCID和 C- PBL- SCID脾细胞对靶细胞有一定的杀伤力 ,B- PBL- SCID的血清能与靶细胞产生凝集反应 .结论  SCID是建立肿瘤模型和肿瘤免疫研究的良好动物 ;肝癌细胞在小鼠体内的成瘤率与 T。 AIM To explore the immunoreconstitution of SCID mice and the affection of immunity on hepatocarcinomacell growth in vivo . METHODS The human hepatocarcinoma cells cultured in vitro were inoculated into the limbs of SCID mice subcutaneously and the immunoreconstituted SCID mice with BALB/c and CBA/N mice peripheral blood lymphocytes (B PBL SCID and C PBL SCID mice) and their growth situations were monitored. The spleen cells of these mice were harvested in 5 weeks after inoculation and examined for direct cytotoxicity to the hepatocarcinoma cells (target cells). The immunoglobulin concentration of B PBL SCID mice was surveyed and the serum agglutination of B PBL SCID mice was tested. RESULTS The successfully reconstituted immunity in SCID mice embodied the rejection characteristics of BALB/c and CBA/N mice to the hepatocarcinoma cells. There was 100% carcinoma growth in SCID and C PBL SCID mice, while the carcinoma of B PBL SCID mice disappeared after certain time. The spleen cells from B PBL SCID and C PBL SCID mice displayed certain cytotoxicity to target cells, but the control group generated no lysis to target cells. The sera immunoglobulin of B PBL SCID mice had the level of g·L -1 and generated agglutination with the target cells. CONCLUSION SCID is good for establishing tumor model and studying tumor immune. Carcinoma grows in the mice is correlated with T and B lymphcytes.
