摘要
目的 观察糖尿病大鼠福辛普利治疗后心肌层粘连蛋白 (L N)、纤维连接蛋白 (FN)及胶原蛋白含量 (CL )的改变对心肌细胞外基质的影响及对心肌的保护作用 .方法 大鼠2 0只建立糖尿病模型后 ,随机分为福辛普利治疗组 (福辛普利组 )及糖尿病未治疗组 (未治疗组 ) ,每组 10只 .福辛普利组以福辛普利 (15 mg· kg- 1 · d- 1 )用水溶解后 ig,未治疗组以等体积水 ig.另取 10只正常大鼠作为空白对照 (正常组 ) .实验周期 5 wk.结果 经图像分析显示心肌组织 L N的吸光度 A值在正常组、福辛普利组及未治疗组分别为 0 .12 5 1± 0 .0 0 35 ,0 .1337± 0 .0 0 98和 0 .2 36 4± 0 .0 136 .FN的 A值在正常组、福辛普利组及未治疗组分别为 0 .0 5 81± 0 .0 0 42 ,0 .0 86 6± 0 .0 0 5 4和 0 .140 4± 0 .0 0 96 .CL 的 A值在正常组、福辛普利组及未治疗组分别 0 .12 0 0± 0 .0 10 4,0 .310 8± 0 .0 2 12和0 .5 114± 0 .0 42 0 .未治疗组 L N,FN和 CL 的表达均明显高于正常组及福辛普利组 ,差异有显著性意义 (P<0 .0 1) .结论 福辛普利虽不能降低血糖 ,然而通过抑制心肌局部肾素 -血管紧张素系统 ,可抑制心肌 L N,FN和 CL的表达 ,明显减轻糖尿病大鼠心肌细胞外基质增生 。
AIM To study the effects of ACEI-fosinopril on the contents of myocardial laminin (LN), fibronectin (FN) and collagen (CL), and its protective effect on myocardium and probable mechanism in diabetic rats. METHODS A total of 30 SD rats were enrolled in the study, of which, 10 were taken as normal controls, others, as models of diabetes by injecting STZ, were equally divided into fosinopril group and untreated group. Myocardial LN, FN and CL were observed in all three groups after 5 wk. RESULTS Image analysis showed that the absorbance ( A ) value of LN in myocardial tissue in control group, fosinopril group and untreated group was 0.1251±0.0035, 0.1337±0.0098 and 0.2364 ±0.0136, respectively. The A value of FN in the three groups was 0.0581±0.0042, 0.0866±0.0054 and 0.1404±0.0096, respectively. The A value of CL in the three groups was 0.1200±0.0104, 0.3108± 0.0212 and 0.5114± 0.0420 , respectively. The values of the three parameters in untreated group were all significantly higher than that in other two groups ( P <0.01). CONCLUSION Fosinopril can not decrease the blood sugar, by inhibiting myocardial renin angiotensin system and expression of myocardial LN, FN and CL, it can significantly decrease the proliferation of myocardial extracellular matrix and relieve the myocardial injury.
