摘要
目的 了解明胶酶A在胶质细胞瘤发展中的作用。方法 取36例脑胶质细胞瘤患者的肿瘤组织及正常脑组织,其中Ⅰ~Ⅱ胶质瘤13例,Ⅲ~Ⅳ级胶质瘤23例。提取组织总RNA;用逆转录共扩增定量PCR方法测定明胶酶A(MMP2)基因表达水平。结果 Ⅲ~Ⅳ级胶质瘤明胶酶A基因表达水平(1.41±0.43)明显高于Ⅰ~Ⅱ级胶质瘤(0.54±0.07)及正常脑组织(0.29±0.04)。胶质瘤MMP2基因表达水平与肿瘤大小无关,而与肿瘤分化程度有关。结论 明胶酶A在脑胶质瘤的进展中可能起重要作用。
Objective To investigate the role of gelatinase A in the progression of human gliomas. Methods Twenty-three patients with grade Ⅲ -Ⅳ of gliomas and 13 patients with grade Ⅰ - Ⅱ of gliomas were selected for the evaluation of gelatinase A (MMP2) gene expression levels, and the normal brain tissues obtained during the operation served as control. The total RNA in the gliomas and the normal brain tissues was isolated and MMP2 gene expression levels were measured by the techniques of reverse transcription polymerase chain reaction. Results MMP2 gene expression level in grade Ⅲ-Ⅳ glioma tissues (1.41±0.43) was significantly higher (P<0.01) than those in grade Ⅰ-Ⅱ glioma(0.54±0.07) and the normal brain tissues (0.29±0.04). MMP2 gene expression levels had strong correlation with the degree of histological differentiation of the tumor but not with the tumor size. Conclusion Gelatinase A may play an important role in the progression of gliomas.
