摘要
目的 探讨人精子甘露糖受体 (MR)的表达与精子所处生理状态的关系。 方法 对人精子进行体外获能和诱导顶体反应处理 ,以 FITC- DMA为探针检测获能前后和诱导顶体反应精子的 MR表达率 ,以 CTC染色监控精子的生理状态 ;对 MR表达的分类百分率与 CTC染色的精子分类百分率作线性回归分析。 结果 MR- 、 型表达率变化均与 CTC监控的精子顶体反应率变化高度正相关 ,MR- 型与获能负相关。 结论 MR的表达反映了精子的顶体反应状况而非获能状况 。
Objective To investigate the relationship between the expression of mannose ligand receptor(MR) and the physiological state of human spermatozoa in fertilization. Methods Fertile human spermatozoa were capacitated in vitro and induced to appear acrosome reaction. The suspensions of sperm before/after capacitation and acrosome reaction were labelled with FITC conjugated D mannosylated albumin(DMA) to detect the MR expression and stained by chlortetracycline(CTC) to monitor the sperm physiological state. Linear regression analysis was used to see their relationship. Results The expression rates of both pattern Ⅱ and Ⅲ MR were intensively correlated to the rate of acrosome reaction of sperm as monitored by CTC, while the expression rate of pattern Ⅱ was negatively correlated to the rate of capacitated sperm showed in CTC staining. Conclusion The expression of MR reflects acrosome reaction but not capacitation in human sperm. The expression of MR is not a marker of capacitation but a companion phenomenon of some sperms with acrosome reaction.
