干细胞白血病基因在急性淋巴细胞白血病的表达与预后

摘要 目的 探讨干细胞白血病 (SCL)基因在急性淋巴细胞白血病 (AL L)中的表达情况及临床意义。 方法 采用 RT- PCR技术检测 SCL 及其融合基因 SIL- SCL 在 AL L 细胞系和患者的表达情况 ,并观察其临床预后关系。 结果  (1) SCL 基因在 AL L 的表达率为 5 1.2 % ,SIL- SCL 基因仅在 T- AL L 中表达 ,占 10 .5 %。(2 ) SCL 阳性表达的 AL L 患者完全缓解率和生存率减低。 结论  (1) SCL 基因对 AL L 的发生有一定作用 ,其阳性表达是 AL L预后不良因素之一。 (2 ) SCL 异常激活的方式以过量表达常见 ,基因重排较少见 。 Objective To illuminate the expression of stem cell leukemia(SCL) gene in acute lymphoblastic leukemia(ALL) and clinical implications. Methods The expression of SCL gene and its fusion gene SIL SCL were detected in cell lines and in ALL patients by RT PCR. The prognosis of the patients was observed. Results (1)The expression rate of SCL gene in ALL was 51.2%, but the expression rate of SIL SCL gene only expressed 10.5% in T ALL. (2)The patients with SCL gene expression had a significantly lower complete remission(CR) rate and poorer survival. Conclusion (1)SCL gene plays certain role in leukemogenesis of ALL. (2)Excessive expression is the common pattern of SCL gene abnormal activation in ALL, and the gene rearrangement is less, and the chromosomal translocation is rare. (3)The SCL gene expression is one of the poor prognostic factors in ALL.
