摘要 目的 探讨 P2 7kip1 蛋白在甲状腺乳头状癌 (PTC)组织中定量表达的临床意义及其与 p5 3的关系。 方法 应用 SP免疫组织化学方法和图像分析技术检测 P2 7kip1 和 P5 3蛋白的表达。 结果 甲状腺滤泡性腺瘤(TFA)、各种不同临床病理分期的 PTC的 P2 7kip1 蛋白阳性细胞的平均积分光密度值 (AODV )依次递减 ,具有统计学意义 (P<0 .0 1) ,而单位面积阳性细胞个数 (NPA )在 TFA与 PTC( 期 )、PTC( 期 )与 PTC( ~ 期 )中差别不显著 (P>0 .0 5 )。在 PTC中 ,p5 3表达阴性组的 P2 7kip1 蛋白的 AODV显著高于 p5 3表达阳性组 (P<0 .0 1)。 结论  P2 7kip1 在 PTC发生和发展过程中发挥重要的作用 ,其蛋白水平和 /或结合 P5 Objective To explore the immunohistochemical quantitative expression of P27 kip1 protein and its relation with p53 in papillary thyroid carcinoma. Methods Microwave SP immunohistochemical technique together with computerized image analysis was used to detect the expression of P27 kip1 and p53 protein. Results The differences of average optic density value(AODV) of P27 kip1 between follicular thyroid adenomas and papillary thyroid carcinomas of different clinicopathological stages(stage Ⅰ, Ⅱ, Ⅲ~Ⅳ) were statistically significant(P<0 01). The level of AODV in four groups of tumor with different clinicopathological stages decreased in turn. Moreover, AODV of P27 kip1 in papillary thyroid carcinomas with negative mutant p53 expression was significantly higher than that with positive p53 expression(P<0 01). Conclusion P27 kip1 plays an important inhibitory role in papillary thyroid cancer carcinogenesis and its development. Its protein expression level with the status of p53 protein expression could serve as a parameter with great value in clinical prognosis. Furthermore, it may provide a strong evidence for the selection of cases with high risk of potential carcinogenesis. Meanwhile, computerized image analysis technique facilitates the evaluation to be more objective and accurate.
