摘要
目的 :研究海星甾醇 C0 1对实验性心律失常的拮抗作用。方法 :麻醉大鼠股静脉快速 iv 5 % Ca Cl2诱发室颤 (VF) ,记录大鼠的 VF发生率和死亡数 ;小鼠尾静脉 iv Ca Cl2 -Ach混合液 (10 ml/ kg)诱发小鼠房颤(扑 ) ,记录房颤 (扑 )的发生率 ;麻醉大鼠冠脉结扎再灌注诱发心律失常 ,记录结扎 5 min和再灌期各时间点的心律失常发生率 ,并比较 ST段的变化 ;电刺激致家兔室颤 ,记录室颤阈值。结果 :海星甾醇 C0 1显著降低了 Ca Cl2诱发 VF的发生率 ,抑制小鼠房颤 (扑 )的发生 ,对大鼠结扎期和再灌期各个时间点都有显著的抗心律失常的作用 ,减轻结扎造成的心肌缺血及再灌所致的心肌损伤 ,中高剂量组可显著提高家兔室颤阈值 ,降低 VF的发生。结论 :C0 1能对抗多个室性和房性心律失常的实验模型 ,提示 C0 1具有广泛的抗心律失常作用。
AIM The purpose is to study the effect of modified starfish sterol (C01) on experimental arrhythmia. METHODS Anesthetized male rats were injected quickly 5% CaCl\-2 through thigh vein to induce arrhythmia. The percent age of appearance of ventricular fibrillation and number of death were recorded. Mice were injected CaCl 2 Ach through tail vein to induce atria fibrillation. The percent age of appearing of atria fibrillation was recorded. Experimental arrhythmia was induced by ligating and reperfusing the rat′s left coranary artery, recording arrhythmic percent age of appearance and comparing value of ST at the different time. Rabbits were stimulated by electricity to induce ventricular fibrillation, recording VFT. RESULTS C01 can reduce VT(%) induced by CaCl 2, restraining appearance of atria fibrillation markedly. Also, it showed effects of antiarrhythmia in the period of ligation and reperfusion. Middle and high dose of C01 can heighten value of VFT, reducing happeness of VF. CONCLUSION The effects of antiarrhythmia on ventricle and atria reveal showed that C01 had broad effects of antiarrhythmia.
