摘要
目的探讨急性放射性皮肤溃疡形成早期b-FGF、VEGF及其受体的表达和意义。方法以50Gy60Coγ射线单次局部照射建立急性放射性皮肤溃疡动物模型,以手术方法建立单纯皮肤伤口动物模型,采用免疫组化、原位杂交等方法检测伤口及溃疡组织内b-FGF、VEGF及其受体的表达。结果创伤组伤后5~10d,肉芽组织内b-FGF、VEGF表达呈强阳性;照射后21~28d,放射性溃疡肉芽组织内b-FGF、VEGF表达呈阳性,但较创伤后5~10d明显减弱。结论辐射诱导的b-FGF、VEGF表达减弱与急性放射性皮肤溃疡愈合延迟有关。
Objective To study the changes and significance for b-FGF and VEGF expression in early stage of acute radiation-induced skin ulcers.Methods A model of normal skin wounds was made.Another model was made with 50Gy 60 Co γ-rays .Immunohistochemical staining,in situ hybridization(ISH)were used to detect the expression of b-FGF and VEGF in simple skin wounds a nd acute radiation-in-duced skin ulcers.Results In simple skin wounds group,the expr ession of b-FGF and VEGF was strongly positive in granulation tissues on 5to 10days after injury;while in radi ation group,the expression of b-FGF and VEGF was weaker in granulation ti ssues on 21to 28days after radiation,compared with simple ski n wounds group on 5to 10days after inj ury.Conclusion Radiation-induced reduction of b-F GF and VEGF was relevant to the wound healin g retardation of acute radiation-in duced skin ulcers.
