T淋巴瘤TCR β链独特型DNA疫苗诱发小鼠体液免疫反应

摘要 目的:研究T淋巴瘤TCRβ链独特型DNA疫苗诱发小鼠体液免疫反应。方法:大剂量提取质粒,将BALB/c小鼠随机分为 pcDNA3.1组、pcDNA3.1/TCR Vβ8组、pcDNA3.1/TCR Vβ8+IL-2组、pcDNA3.1/TCRVβ8+CPG+脂质体组和全硫代CPG组共5组(每组6只),双侧股四头肌内注射疫苗免疫,于第0、2、4周各免疫1次,共3次。每次免疫前及免疫开始后至第8周,取鼠血,应用间接免疫荧光法检测小鼠抗体生成情况。结果:pcDNA3.1/TCR Vβ8组、pcDNA3.l/TCR Vβ8+CpG+脂质体组、pcDNA3.1/TCR Vβ8+IL-2组小鼠血清中均产生了特异性抗独特型抗体,抗体滴度在第4周开始增高,第6周时达到高峰。在同一取血时间,pcDNA3.1/TCRVβ8+CPG+脂质体组和 pcDNA3.1/TCR Vβ8+IL-2组小鼠抗体滴度均高于 pcDNA3.l/TCR Vβ8组(P<0.001)。结论:DNA重组质粒pcDNA3.1/TCR Vβ8可诱导小鼠产生特异性抗淋巴瘤细胞的独特型抗体;IL-2、CPG和脂质体增强了 TCR Vβ8基因疫苗诱导的体液免疫反应。 Aim: To study the humoral immunoreaction of rats induced by idiotype DNA vaccine of TCR β chain of T lymphoma . Methods: A large quantity of plasmids were extracted . The BALB/c rats were randomly allocated into 5 groups (6 in each group) : pcDNA3. 1, pcDNA3. 1/ TCR Vβ8 , pcDNA3.1/ TCR Vβ8 + IL-2, pcDNA3. 1/ TCR Vβ8 +CpG+liposome and thiosulfate-CpG. Vaccines were injected in bilateral musculus quadriceps femorises once at the 0,2nd and,4th week, respectively and 3 times in all. The antibody in rat blood was tested by indirect im-muofluorescence method before immunization and 8 weeks after immunization . Results: All the rats in the groups of pcDNA3. 1 /TCR Vβ8, pcDNA3.1/ TCR Vβ8 + IL-2, and pcDNA3.1/ TCR Vβ8 + CpG+liposome produced special antibody against idiotype antigen in serum. The titer of antibody began to rise in the 4th week and reached the peak in the 6th week . The titers of antibody of rats in the groups of pcDNA3.1/ TCR Vβ8 + IL-2 and pcDNA3.1/ TCR Vβ8 + CpG+liposome were both higher than that of the group of pcDNA3. 1 /TCR Vβ8 at the same time (P< 0. 001). Conclusion: It is testified that the recombinant plamsid of pcDNA3. 1 /TCR Vβ8 could induce rats to produce special antibody against idiotype antigen of lymphoma cells. IL-2, CpG and liposome can improve the humoral immunoreaction induced by TCR Vβ8 genes vaccine.
