秋水仙碱及热击与低温诱导对油菜小孢子胚状体成苗率的影响 被引量:36

摘要 甘蓝型冬油菜 F1 杂种分离小孢子直接用 5 0 0 mg/ L秋水仙碱处理 15 h,在 NL N - 13培养液中培养 ,并于 32℃和30℃分别热击暗培养 3d和 7d后转入 2 4℃下培养 ,胚胎发生好 ,5周后产生大量正常胚体。将这些发育优异的胚体移入固体 MS培养基后即进行 10 d低温诱导 (2℃ ) ,常温 (2 4℃ )光下培养能很好地萌发并直接、快速地再生植株 ,其萌发胚率和成苗率分别达到 89.3%和 86 .7%与 6 2 .5 %和 5 5 .4 %。 The effects of colchicine treatment with heat shock and cold induction on embryogenesis and plant regeneration from microspore derived embryos were investigated in the F 1 hybrids of winter oilseed rape( Brassica napus L.). Freshly isolated microspores were immediately treated with 500 mg/L of colchicine for 15 h and incubated in liquid NLN 13 medium in darkness at 32℃ and 30℃ for 3 d and 7 d, respectively. Large amounts of normal embryos were produced after incubated for 5 weeks. After transferred to solid MS medium and an initial period of cold induction (2℃) for 10 d, these embryos germinated well at 24℃ in the light and directly and quickly regenerated plants. The rates of embryo germination and plant regeneration reached 89.3% and 86.7%, and 62.5% and 55.4%, respectively.
