摘要 目的 观察环氧合酶 (COX)的两种同工酶 ,即COX 1和COX 2在单侧输尿管梗阻 (UUO)大鼠术侧肾脏的表达情况。方法 建立左侧输尿管梗阻模型 ,设假手术组为对照。 4周后分别应用逆转录 聚合酶链反应 (RT PCR)和免疫组织化学方法检测COX 1、COX 2的mRNA水平及蛋白质表达 ,并采用放射免疫法测定术侧肾脏的残余尿液中血栓烷素B2 (TXB2 )浓度。结果 COX 1的mRNA和蛋白水平在两组间差异无显著性。病变组大鼠尿液中TXB2 的浓度急剧上升 ,COX 2的基因表达和蛋白质合成大幅增加 ,病理上呈现明显的肾小管间质纤维化。结论 COX Objective To observe the renal expression of cyclooxygenase (COX) in the rats with unilateral ureteral obstruction (UUO). Methods Rats were evenly divided into two groups. Some were subjected to complete ligation of left ureter and the others with sham operation were used as control. Four weeks later, using reverse transcription polymerase chain reaction (RT PCR) method, we examined the mRNA expressions of COX 1 and COX 2 in left kidney of all these rats. The immunohistochemistry analysis was carried out to investigate the protein level of COX 1 and COX 2. The urinary concentration of thromboxane B 2 was determined by radioimmunoassay. Results There was no difference in the values of COX 1 mRNA and protein between these two groups. In group UUO, the urinary concentration of thromboxane B 2 increased substantially; both the gene expression and the protein level of COX 2 were much higher than those in the control group. At the same time,there appeared tubulointerstitial fibrosis in the kidney of rats with UUO. Conclusion COX 2 is involved in the pathogenesis of the injury of UUO.
