摘要
取 10例风湿性心脏病 (风心病 )窦性心律 (窦律 )患者、10例心房纤颤 (房颤 )患者的右心耳组织 ,应用逆转录 -聚合酶链反应技术 ,以 GAPDH为内参照 ,测定心房肌钙调蛋白 m RNA表达水平。结果显示 ,与窦律组相比 ,房颤患者细胞膜 L型 Ca2 + 通道α 1c亚基及肌浆网钙泵的 m RNA表达减低 ,两组间有显著性差异 (P<0 .0 5 ) ,而磷酸受纳蛋白、钙释放通道和细胞膜 Na+ - Ca2 + 交换的 m RNA表达水平无显著性差异。
Right atrial appendages were obtained fro 10 patients with atrial fibrillation and matched with 10 patients with sinus rhythm.Total RNA was isolated and the mRNA expression of proteins influencing the calcium homeostasis were measured by revese transcription polyerase chain reaction and normalized to the mRNA level of glycersldehyde 3 phosphate dehydrogenase.The mRNA level of L type calcium channel α 1c subunit and sarcoplasmic reiculum calcium ATPase were decreased significantly in patients with atrial fibrillation( P<0.05 ).No changes in mRNA expression of Na + Ca 2+ exchanger,phospholamban and ryanodine receptor type 2 were observed in patients with atrial fibrillation compared with those with sinus rhythm.These data suggese that calcium loading may be at least partly responsible for atrial fibrillation.
