摘要
目的 了解卡托普利对动脉粥样斑块中原癌基因 c- m yc和 c- fos m RNA表达的影响及原位逆转录 - PCR(IS-RT- PCR)技术在检测中的应用效果。方法 将 40只雄性日本大耳白兔随机分为对照组、胆固醇组及用药组 ,采用ISRT- PCR技术观察三组原癌基因 c- myc和 c- fos m RNA的表达及分布。结果 (1)胆固醇组的 c- m yc和 c- fosm RNA阳性率明显高于对照组 ,用药组则显著低于胆固醇组 ;(2 )主动脉粥样斑块中 c- m yc、c- fos m RNA呈强阳性反应 ,广泛分布于内膜层 ,阳性信号为核阳性 ;而用药组呈弱阳性反应 ,散在性分布。结论 提示卡托普利能显著抑制 AS斑块中原癌基因 c- m yc和 c- fos m RNA的表达 ,同时亦体现了 ISRT-
Objective To study effect of Captopril on oncogene c-myc and c-fos mRNA expression in atherosclerotic plaques and applicative effect of in situ reverse transcription-PCR (ISRT-PCR) technique upon experiment. Methods 40 male Japan white rabbits were randomly divided into normal control group, cholesterol group and drug group. Using ISRT-PCR, we studied the expression and location of c-myc and c-fos mRNA oncogene in three groups. Result (1) Positive rate of c-myc and c-fos mRNA in cholesterol group was increased as compared with that of normal control group. Positive rate of c-myc and c-fos mRNA in drug group was significantly decreased as compared with that of cholesterol group; (2) Expression of c-myc and c-fos mRNA was strong positive signal in aortal plaques, particularly at the intema layer. Moreover, positive signals of c-myc and c-fos mRNA were mainly located in the nucleus. Expression of c-myc c-fos mRNA in drug group was doubtful positive. Location of signal was divergence. Conclusion Expression of c-myc and c-fos mRNA in atherosclerotic plaques was significantly inhibited by Captopril. Meanwhile, applicative result of ISRT-PCR technique combining cell type with cell amplification was satisfactory.
