摘要
经紫外线诱变处理和药物梯度培养分别获得具有抗药性标记的北京棒杆菌(Co-rynebacterium pekinense AS1.563-1 Ery^R Str^S Hom^-)和黄色短杆菌(Brevibacte-rium flavum AS1.495-1 Str^R Ery^S Leu^-),研究了它们的原生质体形成,再生及融合条件。从检测融合子抗药性标记、营养缺陷型和发酵氨基酸纸层析结果三方面证明棒杆菌和短杆菌的原生质体发生了融合。直接选取法融合率为6.4×10^(-5),间接选取法融合率为5.2×10^(-3).本文对属间微生物原生质体融合作了有意义的探讨。
This paper describes the condition of protoplast formation,regeneration of Corynebacterium pekinense AS1.563 and Brevibacterium flavum AS1.495 and the intergeneric protoplast fusion of these two strains.Frequency ofr the fusion is 6.4×10^(-5)for direct selection and 5.2×10^(-3)for indirect selection.With the resultsof analysing the stability,auxotroph and fingerprint of amino aoid ehromatogram of eight fusants,stable intergeneric fusants have been obtained. Also,this paper discusses some respects to the protoplast fusion between intergeneric microorganisms.
