丙型肝炎病毒体外感染成年树肝细胞形态学的初步研究

Prelimiary study on morphologic changes of the infected cells of adult normal Tupaia hepatocytes in primary culture by hepatitis C virus in vitro.

目的 :探索丙型肝炎病毒 (HCV)体外感染成年树肝细胞形态学的变化。方法 :HCVRNA阳性血清与培养肝细胞共同孵育后 ,收集不同时相标本并用逆转录聚合酶链反应 (RT -PCR)检测细胞内和上清中正负链RNA ,光镜、电镜观察感染细胞形态学变化。结果 :光镜下未发现感染细胞形态学变化 ,电镜下发现胞浆内含有大量的脂肪空泡、多泡体 ,并可见疑似病毒样结构。结论 :丙型肝炎病毒体外不仅能感染树肝细胞 。

Objective:To explore the morphologic changes of the adult normal Tupaia hepatocytes in primary culture by hepatitis C virus in vitro.Methods:After inoculation,the cells were harvested at various phases during the culturing periods.Both the plus and minus strands of HCV RNA in infected cells or supernatant were examined by reverse transcription polymerase chain reaction(RT-PCR).The morphologic changes of the infected cells were observed under light-microscope and electron-microscope respectively.Results:The infected cells didn't display obvious changes under light-microscope;but ultrastructural examination of the infected cells showed a lot of fat droplet,poly_vesiculiforms and minus-vesiculiforms containing nucleated-substance just like virus-like structures Conclusion:HCV not only infects the liver cells in Tupaia,it also could directly lead to cytopathogenesis in vitro.
