CPP32在耐药细胞及药敏肿瘤细胞中的表达及其意义

CPP32 expression and its significance in multidrug-resistant tumor cells and their parent cells

目的探讨在胃癌多药耐药细胞系中CPP32的表达差异及其对耐药细胞和药敏细胞凋亡的可能影响。方法用已建立的成熟的胃癌多药耐药细胞系SGC7901/VCR08,用RT-PCR的方法分别对耐药细胞和药敏细胞中的CPP32mRNA进行扩增、测序,用Westernblotting的方法检测其蛋白表达。结果基因测序表明,耐药细胞和药敏细胞中CPP32的mRNA表达并无差异,且与文献报道序列完全一致。Westernblotting的结果也显示,CPP32在耐药细胞和药敏细胞中的蛋白表达量无显著差异。结论在耐药细胞和药敏细胞中CPP32的基因序列完全一致,蛋白表达水平无显著差异,提示耐药细胞所表现出的凋亡抗性与CPP32的差异无关,其机制可能在CPP32的上游环节。这一发现揭示我们以CPP32为靶目标展开的促使耐药细胞凋亡的研究具有深远的意义。
