一氧化氮对骨肉瘤细胞体外跨膜侵袭能力的影响 被引量:3

Effect of nitric oxide on the invasive ability of human osteosarcoma through a filter membrane:an in vitro study

摘要 目的研究一氧化氮(NO)对体外培养的人骨肉瘤细胞株(OS-732)侵袭能力的影响。方法将含有不同浓度硝普钠(SNP)的骨肉瘤细胞株悬液加入肿瘤体外侵袭模型中,分别计数细胞跨膜侵袭的数量,分析不同浓度SNP组间细胞跨膜侵袭数量的差异及与对照组间的差异。结果随着SNP浓度的增加,人骨肉瘤细胞株细胞穿过膜的数量呈逐渐减少的趋势,变化范围从平均每个视野184.4±12.8降至33.0±9.0,而PBS组平均为每个视野195.8±14.5。经统计分析,除SNP500μmol/L组与其他各组间均存在显著性差异(P<0.05)外,其余各组(包括PBS组)均与非相邻组间存在显著性差异(P<0.05),而与相邻组间无显著性差异(P>0.05)。结论NO可以在一定程度上抑制骨肉瘤细胞的体外跨膜侵袭能力。 Objective To studytheeffectof nitricoxideon thein vitro invasivenessof humanosteosarcomacellline(OS-732).Methods In vitro culturedOS-732cellsuspensionscontainingdifferentconcentrationsof sodiumnitroprusside(SNP)or PBS(controlgroup)wereusedto assessthein vitro invasivenessof thetumorcellsutilizingBoyden'schamberassay.Thetumor cellsgrowingacrossthefiltermembranewerecountedby meansof HEstainingandcomparedbetweenthegroups.Results Withtheincreaseof SNPconcentrationinthecellsuspension,thenumberof tumorcellsgrowingacrossthemembranetended to decrease.Withtheexceptionof thecellsuspensioncontaining500μmol/LSNPthathada cellnumberacrossthemembrane significantlydifferentfromthoseof anyothergroups,significantdifferencewas notobservedbetweenanyadjacentgroups.BetweenthegroupswithSNP concentrationsthatdidnot comeadjacent,however,significantdifferenceswerenoted.Conclusion Nitricoxidecandepresstheinvasivenessof OS-732cellsacrossthemembranein vitro.
