摘要
目的 :寻找从兔面神经中快速分离和纯化许旺氏细胞的方法。方法 :以胰蛋白酶和胶原蛋白酶对神经进行消化 ,找出最佳的消化时间后 ,采用多聚赖氨酸特异性粘附法纯化许旺氏细胞。结果 :神经消化时间以 4 5~ 6 0min时所得到的细胞数量最多 ,经粘附处理后 ,排除了成纤维细胞 ,使许旺氏细胞的纯度明显增加。结论 :采用酶消化法和L 多聚赖氨酸特异性粘附法 ,可在较短时间内获得高纯度的许旺氏细胞。
Objective :to investigate a method to obtain pure Schwann cells from facial nerve rapidly Methods: to find the proper digest time at which the yield of Schwann cell is highest by the use of 0.25% Trypsin(Sigma) and 0.03 % Collagenase(Sigma) in the digest process of the facial nerve and then to use the L polylysine differential adhesion method to exclude fibroblasts. Results: the yield of Schwann cell is highest when the digest process is between 45~60 minutes and the most fibroblasts had been excluded. Conclusion: by the use of trypsin and collagenase in digest process and the L polylysine differential adhesion method highly pure Schwann cell can be obtained in short time.
