摘要
目的 :研究药物中毒性耳聋大鼠耳蜗 4种热休克蛋白 (Heatshock protein,HSP)的表达特点 ,探讨 HSP在听力损伤及修复过程中的作用及意义。方法 :实验组大鼠腹腔注射硫酸庆大霉素造成慢性药物中毒性耳聋 ,取耳蜗作石蜡包埋切片 ,免疫组化 SP法检测 4种 HSP表达。结果 :4种HSP在正常大鼠耳蜗各回均有不同程度的染色 ,主要阳性部位是螺旋神经节、Coti氏器和血管纹 ,螺旋韧带有较弱的表达。耳聋大鼠耳蜗 4种 HSP表达的一般特征同上 ,但 HSP70、HSP60及HSP2 7在各阳性部位的表达均明显增强 ,而 HSP90表达变化不明显。结论 :多种 HSP在大鼠耳蜗组织均有选择性表达 ,HSP可能参与耳聋发病过程 。
Objective: To investigate the expression of four HSPs in cochlea of rats with deafness and to determine their effect and significance in the hearing loss and recovery. Methods: Deafness model of rat was established by intraperitoneal injection of sulfate gentamycine. Cochleas embedded in paraffin were dissected. The expression of HSPs was detected by immunohistochemistry SP. Results: The four HSPs had been stained in the cochlear,especially in the organ of Corti,stria vascularis,and spiral ligand. The general characteristics of deaf rats was the same as normal rats, however, expression of HSP70, HSP60, and HSP27 were significantly enhanced, respectively, whereas HSP90 changed nothing. Conclusion: HSPs are selectively expressed in rat′s cochlea,and HSP70, HSP60 and HSP27 involed in the mechanism of deafness may play a important role in the recovery.
