摘要 目的 观察丝裂霉素 C (m itomycin C,MMC )在青光眼滤过术中单纯巩膜瓣上应用与巩膜瓣上下联合应用对术后成纤维细胞增殖活性的影响。方法 在兔眼实验性小梁切除术中 ,一组用浸泡有 0 .4 g· L- 1 MMC的海绵片 ,置于巩膜瓣上 5 m in,另一组用浓度相同的 MMC置于巩膜瓣上 3min、巩膜瓣下 2 m in。生理盐水做对照。采用 HE染色及增殖细胞核抗原 (proliferation cell nuclear antigen,PCNA)染色 ,观察术后 1、4、7、14及 2 8d滤过泡区的组织学改变及成纤维细胞增殖活性的变化。结果 应用 MMC组 PCNA阳性成纤维细胞在术后第 4天开始增多 ,但细胞数目远较对照组少。随时间延长 ,细胞数目增加不明显 ,14 d后阳性细胞明显减少。瓣上组与瓣上下组间 PCNA阳性细胞数目经统计学处理无显著性差异 ,而且 2组成纤维细胞增殖的动态变化趋势相同。结论  MMC瓣上与瓣上下联合应用的抗纤维增殖效果相同。 Objective To investigate the effects of mitomycin C (MMC) on the cell replication activity in different applied methods following experimental filtration surgery.Methods During trabeculectomy in rabbits eyes ,MMC (0.4g·L -1 ) was applied on episcleral for 5 minutes before preparations of a half-thickness scleral flap in one group; MMC was applied on episcleral for 3 minutes combined with in scleral bed for 2 minutes in another group. The control group underwent the same treatment but with balanced salt solution. At 1?4?7?14 and 28 days after surgery, we observed histological changes and expression of proliferation cell nuclear antigen in the filtering area. Results In the MMC-treated eyes, the immunoreactive cells began to increase 4 days after surgery. But the number of immunoreactive cells in the treated eyes was much lower than that in controls. There was no significant difference between the two MMC-treated groups.Conclusion MMC suppresses the proliferation of fibroblasts in the filtering site, and no differences are found between the two MMC-treated groups .
