肿瘤细胞中两种TNFR的表达及新型rhTNF对肿瘤细胞生长的影响 被引量:4

摘要 目的 探讨TNFRp55和TNFRp75在各种肿瘤组织中的表达与新型rhTNF对肿瘤细胞生长的抑制作用的关系。方法应用免疫组织化学染色法,观察120例肿瘤组织和10株体外培养的肿瘤细胞中,TNFRp55和TNFRp75的表达及分布。同时应用结晶紫摄入法,观察nrhTNF对肿瘤细胞生长的抑制。结果 在肿瘤组织和肿瘤细胞中,TNFRp55的阳性表达率分别为82.5%和99%,TMFRp75分别为 84.2%和98.8%,不同类型肿瘤间的表达无明显差异(P>0.05).nrhTNF可抑制SW480、SGC7901、8910SMMC7721和BEL7402这5株肿瘤细胞生长,但是不能抑制另外5株肿瘤细胞A549、PLA801、Hela、Eca109和GRC-l的生长。结论TNFR广泛存在于各种肿瘤细胞上,nrhTNF抑制肿瘤细胞生长的程度与TNFRp55的表达率无相关性。 Aim To investigate the relationship between the expres- sion of TNFRp55 and TNFRp75 in tumor tissues and tumor cell lines in vitro and the supression of novel rhTNF on these tumor cell lines growth. Methods Expressions of TNFRp55 and TNThp75 in l20 tumor tissues and 10 tumor cell lines in vitro was evaluated by immu- nohistochemical staining. The suppression of nrhTNF on tumor cell lines growth was detected by using crystal violet staining. Results In these tumor tissues and tumor cells, the positive rates of TNFRp55 expression were 82. 5% and 99% respectively, whereas those of TNFRp75 expression was 84. 2% and 98. 8% respectively. There were no significant difference among different types of tumors(p > 0. 05). nrhTNF could inhibite SW480, SGC79O1, 8910, SMMC772I and BEL7402 cell lines growth and had no effect on A549, PLA8O1, Hela, EcalO9 and GRC- 1 cell lines. Conclusion TNFRs exist on different tumor cells. There are no correlation between nrhTNF inhibition on tumor cell lines growth and the TNFRp55 expression on these tumor cells.
