摘要 目的 :探讨细胞周期素依赖性激酶 2 (Cdk2 )及其抑制因子p2 7Kip1与胃癌生物学行为的关系。方法 :采用免疫组织化学S P法对 6 0例胃癌组织原发灶和淋巴结转移灶进行Cdk2、p2 7Kip1免疫组化染色 ,腹水或腹腔冲洗液行脱落细胞学检查。结果 :Cdk2阳性表达率 5 6 .6 7% (34/ 6 0 ) ,阳性细胞定位于细胞核和细胞浆 ,以Ⅲ、Ⅳ期表达率较高 (P <0 .0 1) ,与胃癌大体类型及浆膜分型关系密切 (P <0 .0 1) ;p2 7Kip1阳性表达率为 4 3.33% (2 6 / 6 0 ) ,阳性细胞定位于细胞浆 ,在高分化及Ⅰ、Ⅱ期胃癌高表达 (P <0 .0 5 ) ,且与淋巴结转移密切相关 (P <0 .0 1) ;Cdk2、p2 7Kip1同时阳性表达 19例 ,占 31.6 7% (19/ 6 0 )。结论 :Cdk2与胃癌浸润深度、淋巴结转移呈正相关 ,p2 7Kip1表达与胃癌浸润深度。 Objective: Our aim was to evaluate the relationship between cyclin dependent kinase 2 (Cdk2), p27 Kip1 expression and biological behavior, lymph node (LN) metastasis and peritoneal metastasis of gastric cancer. Methods: The expressions of Cdk2 and p27 Kip1 were investigated by using immunohistochemical method (Streptavidin Peroxidase, S P) in 60 cases of formalin fixed, paraffin embedded primary and metastatic foci of gastric cancer. Systematic pathological examinations and exfoliated cancer cells (ECC) were performed. Results: The positive rate of Cdk2 expression was 56.67% (34/60) in all the 60 cases with gastric cancer. Its protein was located mainly in both the nuclei and the cytoplasm, the positive expression of Cdk2 was found mainly in cases at stage Ⅲand Ⅳof gastric cancer and also correlated with Borrmann typing and peritoneal metastasis of gastric cancer (P<0.01); The positive rate of p27 Kip1 expression was 43.33% (26/60), its protein was located mainly in the cytoplasm. There was high p27 Kip1 expression in gastric cancer with differentiated,negative LN metastasis and stageⅠandⅡ(P<0.05). The expression rate of Cdk2 and p27 Kip1 was 31.67% (19/60). Conclusion: The expression of Cdk2 was correlated positively with the depth of invasion and LN metastasis of gastric cancer; whereas the relationship between p27 Kip1 expression and depth of invasion and LN metastasis of gastric cancer was correlated negatively.
