摘要
小麦经200mmol NaCl溶液培养3天后,采用改进的焦锑酸钾方法对叶肉细胞中Na^+及Cl^-进行超微结构定位。电镜观察及电子探针X-射线显微分析表明,Cl^-主要分布在细胞间隙、细胞壁及细胞质膜中。用电子探针X^-射线能谱仪在这些部位中未探测出Na^+,提示Cl^-比Na^+更多地进入小麦的叶肉细胞。此外,在叶肉细胞的细胞核、线粒体及叶绿体中也可见到离子沉淀颗粒。经氯化钠溶液培养的小麦幼苗,其叶肉细胞的叶绿体、线粒体的超微结构受损,植株生长受到抑制。
The distribution of Na^+ and Cl^- in wheat leaf mesophyll cells from plants grown in the solution culture with 200m tool NaCl was studied by means of electron microscopy and X-ray microanalysis.Under electron microscope more antimonate precipitates were fcund deposited in the plazmalemma, cell walls, triangular schizogenous spaces, nucleus, mitochondria and chloroplastids of wheat leaf mesophyll cells from the plants grown in the solution culture with NaCl than from those grown in double-distilled water. The X-ray microanalysis showed that Cl^- had occurred largely in the plazmalemma, cell walls and triangular schizogenous spaces, and Na^+ hadn't been detected in these sites by means of X-ray microanalysis. The results suggested that fewer Na^+ reached wheat leaves than Cl^- did.The ultrastructure of chloroplastids and mitochondriain mesophyllous cells of wheat grown in the solution culture with 200m tool NaCl was damaged.In addition, the growth of wheat plants was checked in the solution of NaCl.
