摘要
目的 : 比较自发性高血压大鼠 (SHR)和WKY大鼠心肌核因子 κB的表达以及厄贝沙坦和咪达普利对SHR心肌核因子 -κB的影响。方法 将 2 1只雄性自发性高血压大鼠 (SHR)随机分成 3组 ,每组 7只。其中 2组分别灌喂厄贝沙坦 50mg/kg/d、和咪达普利 3mg/kg/d ;另一对照组给正常饮水 ,并与雄性同周龄WistarKyoto大鼠 (WKY) 6只比较。共 1 3周 ,免疫组织化学方法检测四组大鼠心肌核因子 -κB的表达。结果 SHR对照组心肌核因子 κB表达明显高于WKY组 ;用厄贝沙坦和咪达普利后核因子 κB表达下降、血压均显著抑制 ,明显低于高血压对照组 (P <0 0 1 ) ,两治疗组的心脏湿重 /体重 (HW/BW)也显著低于对照组 (P <0 0 1 )。结论 厄贝沙坦和咪达普利在抑制血压和左室肥厚的同时也明显抑制心肌核因子 κB的激活 ,提示AT1 拮抗剂和ACEI对核因子
Objective To explore the suppressing effect of Irbesartan and Imidapril on activation of Nuclear Transcription Factor κB(NF κB) in cardiomyocte of spontaneously hypertensive rats. Methods Twenty one SHR were divided randomizedly into 3 groups of 7 rats each. Two groups were fed with irbesartan of 50 mg/kg, imidaprill of 3 mg/kg per day for 13 weeks respectively,while the remained group (SHR control group)and 6 Wistar Kyoto rats (WKY) were given normal water only for equal time. Nuclear Transcription FactorκB activation were studied by immunohistochemistery. Results It was shown that NF κB expressiong of SHR control group was higher than WKY groups, and it was shown markly lower expression in irbesartan group or imidapril than that in SHR control group; and that blood pressure and the value of heart weight/body weight were suppressed markedly ( P <0 01) in both groups fed with irbesartan and with imidapril compared with SHR control group. Conclusions Both irbesartan and imidapril have the suppressing effects of NF κB activation while suppressing LVH and BP. It suggest that NF κB inhibition will ameliorate target organ damage in hypertension patients.
